The long-term goals of this proposal are to determine if dynamic changes in the size of CNS synapses occur during development, and if so, to investigate the mechanisms underlying this structural plasticity. Furthermore, a major goal is to determine if structural plasticity of CNS synapses is unique to the CNS or has a general significance in the brain. The proposed experiments make use of the chick ciliary ganglion (CG), which provides a unique model system in which to address these questions. The most striking feature of the CG is its topography of connections with the brain, which exhibits dramatic structural plasticity during development. Thus, the CG provides an opportunity to test how structural plasticity occurs at CNS synapses in general. Specifically, I will determine: (1) whether a developmental increase in synapse size occurs in the chick CG; (2) if so, what regulates synapse growth; and (3) whether this type of synapse growth is unique to the CNS or has a general significance in the brain. The proposed experiments will use a combination of ultrastructural, immunocytochemical, and tract tracing techniques to explore these questions. Two hypotheses will be tested: (1) that the increase in synapse size is due to an increase in the number of synapses; and (2) that the new synapses are formed or enlarged from existing synaptic spines. These experiments will provide a significant contribution to the understanding of the morphological development of CNS synapses and may provide insight into mechanisms of synapse formation and synapse replacement in the mature brain.However, many small wireless broadband products do not incorporate an active device-to-network communication function and/or may not support dynamic device-to-network communication.
